摘 要:目的探讨糖尿病(DM)大鼠认知功能与额叶皮质和海马神经元凋亡的关系。方法电迷宫法测试28只DM大鼠(DM组)和30只正常大鼠(NC组)学习能力,并检测额叶皮质和海马发生凋亡的神经元数目和bcl-2、Bax基因表达。结果DM组大鼠学习能力明显下降,额叶皮质和海马细胞凋亡数高于NC组,bcl-2表达减少,而Bax表达增加。DM组大鼠达到学会标准所需训练次数与额叶皮质和海马细胞凋亡数呈正相关,而NC组大鼠两者无相关性。结论高血糖状态下额叶皮质与海马凋亡调节基因表达改变引起的细胞凋亡增加可能与糖尿病认知功能障碍有关。Objective To discuss the relationship between cognition function and apoptosis status in the cortex of frontal lobe and hippocampus of diabetic rats. Methods Learning ability of 28 diabetic rats and 30 normal rats were measured by electric maze after 12 weeks. Then the rats were executed to obtain the brain tissue to estimate the apoptosis cells and expression of bcl-2 and Bax genes in the cortex of frontal lobe and hippocampus. Results The learning ability of diabetic rats were worse than that of control group(P〈0. 001). Apoptosis cells in cortex of frontal lobe and hippoeampus were more in DM group than in NC group. The expression of bcl-2 gene was reduced while the Bax gene was increased in diabetic rats versus control rats. The training times for rats to reach the learning standard were correlated positively with the number of apoptosis cells in cortex of frontal lobe and hippocampus in diabetic rats (r=0. 410, P 〈0. 05), but not in control group(r=0. 263, P〉0.05). Conclusions The increased apoptosis may contribute to cognition dysfunction in diabetes mellitus.
