摘  要:目的观察添加负载银离子的羟基磷灰石/二氧化钛纳米复合材料(nAg-HA/TiO2)对成骨细胞超微结构及细胞活性的影响。方法将成骨细胞分别在添加了不同浓度(10μg/mL^500μg/mL)的nHA或nAg-HA/TiO2的培养基中培养2、6、8、24、72及120h,通过透射电镜观察、细胞计数及MTT法检测细胞超微结构、细胞增殖及线粒体活性,并计算单位细胞呼吸率。结果与空白对照组相比,纳米材料组细胞增殖及总体线粒体活性均受到抑制(P<0.05),且纳米材料浓度越高,对细胞的抑制作用越显著。但纳米材料组细胞呼吸率即单位细胞线粒体活性较高(P<0.05)。纳米材料对细胞超微结构无明显影响;同浓度nAg-HA/TiO2组与nHA组之间各项检测结果的差异无统计学意义(P>0.05)。结论nAg-HA/TiO的成骨细胞相容性与nHA近乎相同。Objective Osteoblasts behaviors were investigated when cultured in the presence of silverhydroxyapatite/titania nanoparticles (nAg-HA/TiO2) in vitro. Methods Rat calvaria osteoblasts were cultured in the presence of 10 μg/mL-500 μg/mL nHA or nAg-HA/TiO2 for 2 h, 6 h, 8 h, 24 h, 72 h and 120 h. Cell proliferation, vitality and ultrastrueture were tested using cell counting chamber, MTT assay, and transmission electron microscope (TEM). Results Both cell proliferation and vitality were restricted by addition of nanopartieles, while the respiration rates as an index for unit cell viability were enhanced, nAg-HA/TiO2 at low concentration showed superiority. No remarkable ultrastructural changes were observed in the osteoblasts exposed to nanopartieles. The differences in cell proliferation and cell vitality between nAg-HA/TiO2 group and nHA group at the same concentration were insignificant. Conclusion nAg-HA/TiO2 restricted cells proliferation, but triggered unit cell viability. Cytoeompatibility of nAg-HA/TiO2 is almost the same as that of nHA's.
