摘 要:目的研究胰岛素对糖尿病模型肾小管骨桥蛋白(OPN)表达的影响。方法腹腔注射链脲佐菌素(65mg/kg体重)复制大鼠糖尿病模型。治疗组每日注射胰岛素(3U/d),共12w。用不同浓度葡萄糖处理MDCK细胞后,加入胰岛素(100mU/L)。免疫组织化学和Western blot检测OPN表达的变化。结果与正常对照组相比,糖尿病组肾小管OPN表达显著升高,胰岛素治疗组肾小管OPN表达较糖尿病组则明显下降。胰岛素明显降低高糖诱导肾小管上皮细胞OPN的表达。结论胰岛素可明显降低糖尿病模型肾小管OPN的表达。Objective To study the effect of insulin on the expression of osteopontin (OPN) in the renal tubule of diabetic models. Methods MDCK was treated with series concentration of D-glucose, before adding insulin (100 mU/L). Diabetes was induced by intraperitoneal injection of streptozotocin (STZ 65 mg/kg body wt). Insulin was injected 3U/d a day for 12 weeks. The expression of OPN was analyzed in renal tissue by immunohistochemistry and western-blot methods respectively. Results The OPN expression of diabetic model group was significantly increased compared with that of the control group. The treated group of insulin was obviously reduced compared with the model group and still higher than the control group. The expression of OPN induced by high glucose was decreased significantly in MDCK by insulin. Conclusions Insulin can reduce the expression of OPN in the renal tubule of diabetic models significantly.
