白术、茯苓、白术茯苓汤对脾虚大鼠VIP及VIPR2的基因表达的影响

摘　　要：目的:研究白术、茯苓及白术茯苓汤对血管活性肠肽(VIP)及其VIPR2基因表达的影响。方法:运用大黄厚朴枳实加饥饱失常法复制脾气虚证的动物模型,采用放射免疫及RT-PCR等方法,观察白术、茯苓、白术茯苓汤对脾气虚大鼠血浆及结肠组织VIP的影响。结果:(1)与模型组比较,白术茯苓汤组能下调血浆及结肠组织VIP含量(P<0.05),茯苓组呈下调趋势,白术组则不明显;(2)与模型组比较,三组方药均可使脾虚大鼠VIPR2的基因表达量下降(P<0.05)。结论:(1)白术茯苓汤组下调脾气虚大鼠VIP含量优于单味药,表明中药配伍运用的必要性和科学性;(2)三组方药均能降低脾虚大鼠VIPR2的基因表达量,可能与白术、茯苓及白术茯苓汤对VIP的作用靶点有异。Objective： To study the effects of Rhizome Atractyiodis Macrocephalae （RAM）, Poris, Baizhufuling decoction on vasoactive intestinal peptide （VIP） and the gene VIPR2 expression of Spleen- Qi deficiency rats. Methods： The rat model of Spleen- Qi deficiency was made with the use of Radix et Rhizoma Rhei, magnolia bark, Fructus Aurantii Immaturus, and irregular diet, With the method of RIA and RT- PCR, we observed the effect of RAM, Poria, Baizhufuling decoction on VIP in plasma and colon of Spleen- Qi deficiency rats. Results ： （ 1 ） Compared with model group, the level of VIP in plasma and colon was lowered by Baizhufuling decoction group （ P 〈 0.05）, and Poria group had a tendency of lowering VIP content. But there was no obvious decrease of VIP content in RAM group. （2） Compared with model group, the three groups could decline the gene VIPR2 expression of Spleen deficiency rats （ P 〈 0.05 ） . Conclusion： （1 ） Baizhufuling decoction declines VIP content in Spleen - Qi deficiency rats better than single drug does. It demonstrates the necessity and science of making use of TCM compatibility; （2 ） RAM, Poris, Baizhufuling decoction can reduce the gene VIPR2 expression of Spleen deficiency rats. This might be related to the different targets that RAM, Poris, Baizhufuling decoction function.
