摘 要:目的利用大骨节病病区粮食分离出产毒镰刀菌,生物合成T-2粗毒素。方法从大骨节病病区玉米粮样中分离出产毒镰刀菌,经纯化培养后,接种于无菌玉米培养基,光镜下根据菌落特点和菌体形态、分生孢子的特点来鉴别菌种.而培养产物经有机溶剂提取、减压浓缩和硅胶柱层析纯化所得浓缩液,用薄层层析法和高效液相色谱法鉴定毒素和产毒量。结果从病区粮样中分离出的产毒菌株为三线镰刀菌,将其接种于玉米培养基,可得T-2粗毒素约250mg/kg。结论大骨节病病区存在着镰刀菌的污染,并由此带来T-2毒素污染。T-2毒素生物合成及提取方法有可行性。其成本较低,适于实验室研究。Objective To isolate Fusarium species from Kashin-Beck disease(KBD) area and biosynthesize crude T-2 toxin. Methods T-2 toxin-producing Fnsarium was isolated from corns produced in KBD area and purified. The purified fungi were identified according to the traits of colony, appearance of thallus and characters of conidium and then were cultivated in sterile corn culture media. After extraction with organic solvent and purification by silica gel chromatography column, the quality and quantity of the toxin in the extracts were estimated by thin-layer chromatography and high-performance liquid chromatography. Results The toxin-producing strain was Fusarium tricinctum. The corn culture media inoculated with this strain produced about 250 mg of crude T-2 toxin per kg. Conclusions This experiment has indirectly further confirmed pollution of T-2 toxin-producing Fusarium existed in KBD area. The T-2 toxin-producing method is feasible and low costed, and suitable for laboratory experiments.
