摘  要:目的优化鬼臼毒素固体脂质纳米粒配方及工艺条件。方法采用正交设计方法对制剂配方及工艺条件进行实验,以表面活性剂含量及表面活性剂成分,固体脂质的含量,搅拌速度为4个因素,固体脂质纳米粒粒径大小为考察指标,筛选出鬼臼毒素固体脂质纳米粒最佳的制剂量配方,并进行形态学和稳定性观察。结果通过对各配方的综合评价进行统计分析研究得出表面活性剂含量为5%,主要成分为Brij78,固体脂质含量为1%,搅拌速度1000 r/min为最佳制备条件,鬼臼毒素含量为0.5%。结论根据正交实验结果制备的鬼臼毒素固体脂质纳米粒在粒径方面符合要求,且在稳定性、外观方面均较理想。Objective To select the optimum condition of the podophyllotoxin solid lipid nanopartcles (POD-SLN) preparation. Methods The optimization of POD-SLN preperation was performed based on the orthogonal esperimental design. In this study, the particle size of POD-SLN were taken as the index and the influenece of factors such as concentration of surfaetant, matrix lipid,the composition of surfactant and agitation velocity were evaluated. Results The best formulation of POD -SLN were 5% surfaetant (Brij78), 1% matrix lipid and agitation velocity was 1 000 r/min, in which podophyllotoxin was 0.5%. Conclusion The POD - SLN designed according to orthogonal experiment was coincidenced with the requirements in aspect of the particle size and it has been proved to be stable, reasonable and scientific.
