摘  要:采用低温-胃蛋白酶法从新鲜猪皮中提取胶原。通过十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)结合紫外、红外光谱分析证明所提取的胶原为I型活性胶原,即胶原分子量约为32万道尔顿,组成为(1α)22α,且具有完整的三股螺旋结构;利用天然胶原的特性降解酶细菌胶原酶进行低温降解,得到分子量在3万到20万道尔顿之间的降解产物,降解产物红外光谱证明大部分仍然保持三股螺旋结构,并与市售的胶原多肽(失活)的红外光谱图比较,充分体现了此降解方案的优越性。Collagen was extracted from fresh pig-skins by pepsin digestion at low temperature. SDS-PAGE, UV and FTIR analysis suggest that the extraction products were active type Ⅰ collagens, the molecule mass of which is about 320k dalton. It is consisted of (α1)2α2, and have the integrated triple helix structure; Collagens were digested by bacterial collagenase which is one of collagen-degrading enzymes, to gain small fragfnents with various molecular weights from 30K to 200K Dalton. FTIR analysis demonstrated that the triple helix structures of them were rather preserved even after fragmentation. What's more, The FTIR image of segment sample was compared with that of the purchase, which sufficiently-reflects advantages of this digesting-scheme.
