摘 要:目的建立测定当归中丁烯苯酞含量的高效液相色谱(HPLC)法。方法样品用甲醇超声提取,外标法定量,色谱柱为ZorbaxSB-C18柱(150mm×4.6mm,5μm),流动相为甲醇-5%异丙醇水溶液(60:40),检测波长为230nm,流速为1mL/min,柱温25℃。结果丁烯苯酞进样量在0.1~1.0μg范围内与峰面积线性关系良好,平均回收率为101.3%,RSD=1.8%(n=6)。结论所用方法简便、准确、重现性好,可用于当归中丁烯苯酞的含量测定。Objective To determine butylidenephthalide in Radix Angelicae Sinensis with RP- HPLC. Methods The sample was extracted with methanol by ultrasonication. The external standard method was used to quantify butylidenephthalide. HPLC separation was carried out in Zorbax SB-C18 column (150 mm ×4. 6 mm, 5μm), and eluted at 1 mL/min with methanol-5% isopropyl alcohol (60:40) as the mobile phase at 25 ℃. The detection wavelength was at 230 nm. Results The linear range was within 0. 1- 1.0 μg for butylidenephthalide. The average recovery rate was 101.3%, and RSD was 1.8% (n=6). Conclusion This method is simple. It can be used to determine butylideneph- thalide in Radix Angelicae Sinensis with satisfactory accuracy and reproducibility.
