摘 要:目的观察老年大鼠与青年大鼠造血功能的差别。方法采用含有15%刺激因子(GM-CSF)的大鼠肺条件培养基进行单层软琼脂法培养大鼠骨髓造血祖细胞,进行定量观察。结果肺条件培养基内刺激因子(GM-CSF)浓度为15%时活性最好。2.5月龄大鼠骨髓,每4×105个有核细胞中形成细胞集落(GM-CFU)为92.5±23.8,而24月龄老年大鼠则为23.1±9.6。以2.5月龄青年大鼠GM-CFU为100%,12月龄大鼠为37%,18月龄大鼠为35.7%,24龄大鼠则为27.2%。结论骨髓造血功能随月龄的增加逐渐减弱,老龄大鼠骨髓造血功能明显下降。Objective To study the difference of haematopoietic function between rats of different months-old age, especially the aged and young rats. Method Bone marrow haematopoietie ancestral cells were cultured in the monolayer soft agar of the lung cell culturing conditioned medium containing 15 % colony stimulating factor ( GM-CSF. ). The cell colonies of karyoeytes per 4 ×10^5 were calculated. Result There were 92.5±23.8 colonies in the young rats of 2.5 month-old, but 23.1±9.6 colonies in the aging rats of 24 month-old. As the GM-CFU of the 2. 5month-old rats was considered as 100% ,it was 37.0% of the 12month rats, 35.7%of the 18 month old rats, and 27.2 of the 24month old rats. Conclusion The haematopoietic function of rats decreased as the age was increased.
