摘 要:目的:建立乌头碱中毒的薄层色谱快速检验方法。方法:样品加乙醚,(NH4)2SO4后调pH到10,分出有机层加盐酸,弃去乙醚层,水层加二氯甲烷和氨水提取,挥干后20μl乙醇定容,10μl点于HPGF254板上,环己烷:乙酸乙酯:二乙胺(8:1:1)饱和上行展开5-7 cm,薄层色谱扫描定性和定量检测。结果:乌头碱线性范围为1-32μg/ml,新乌头碱与次乌头碱均为4-28μg/ml,最小检出浓度0.1μg/ml。结论:本法快速、简便,定性定量准确,可用于乌头碱中毒的快速诊断和乌头碱中毒致死的法医学鉴定。Objective:To develop a rapid qualitative and quantitative analysis method for aconites poisoning by TLCs.Methods:The matrix was extracted twice by ether,(NH4)2SO4,dichlormethane and ammonia water.Evaporated in a water bath and with an air velocity of nitrogen gas,extract was then dissolved with ethanol and analyzed by TLCs.Results:The extraction curves of aconites was linear over the range of 1~32 μg/ml,the extraction curves of new-aconites and hypo-aconites were linear over the range of 4~28 μg/ml.The limit of detection was 0.1 μg/ml.Conclusion:The analysis is more rapid,simpler,more sensitive and more precise.It is a better method for diagnosis of aconites poisoning and forensic identification of aconites poisoning death.
