摘 要:目的:观察N型钙通道在福尔马林诱导的内脏炎症痛中的作用。方法:选用成年健康Wistar大鼠,随机分5组:正常对照组(N);单纯福尔马林直肠致炎组(F);脊髓蛛网膜下腔插管组(IT),插管内注射生理盐水组(NaCl);插管内注射SNX-111组(SNX-111)。记录实验组大鼠的疼痛行为反应,致痛后以15min为一个时间段,共记录8个时间段,计2h,分别计算疼痛分数;福尔马林致痛后30min用原子吸收光谱分析和La^3+阻断技术测定L6—S1-2阶段脊髓细胞内Ca^2+含量。结果:F、IT组与NaCl组8个时间段内疼痛分数无显著性差异(P〉0.05);IT组与F组相比也无明显差异(P〉0.05);SNX-111组与NaCl组在致痛后75min内疼痛分数明显减少(P〈0.01或P〈0.05)。F、IT组与NaCl组在福尔马林致痛后30min脊髓细胞内Ca^2+含量无明显变化(P〉0.05);IT组与F组相比也无明显差异(P〉0.05);SNX-111组与NaCl组相比Ca^2+含量明显减少(P〈0.01)。结论:SNX-111能明显减轻福尔马林内脏炎症痛行为反应,降低脊髓内Ca^2+含量,提示N型钙通道可能在福尔马林致内脏炎症痛感受中起重要作用。Objective: To examine the role of N-type calcium channel in visceral inflammatory pain induced by formalin. Methods: Adult healthy Wistar rats were divided into 5 groups randomly: normal group (N) ; group F: only injected of formalin in the submueosa of rectum; group IT: intrathecal insertion of a PE-10 tube ; group NaCl : intrathecal injection of 0.9% NaCl ; group SNX-111 : intrathecal injection of SNX-111. The numerical score for pain behaviors was recorded every 15 minutes for 2 hours. Pain scores were counted respectively. The intracellular calcium contents of L6 - S1 -2 segments of the spinal at 30 min time point after formalin injection were examined by the technique of atomic absorption spectectomy with La^3 + blockage. Results : There was no significant diffference of pain scores in 8 time points between group F or IT and group NaCl ( P 〉 0.05 ) ; and between group IT and group F (P 〉 0.05 ) ; pain scores of group SNX-111 was decreased significantly in the former 75 minutes than that of group NaCl(P 〈 0.05 or P 〈 0.01 ). There was no significant difference of the intracellular calcium contents at 30 min time point after formalin injection between group F or IT and group NaCI( P 〉0.05 ) ;and between group IT and group F( P 〉0.05 ) ; the intracellular calcium contents of group SNX-111 were decreased significantly than that of group NaCl ( P 〈 0.01 ). Conclusion : SNX-111 can significantly alleviate the reaction of visceral inflammation induced by formalin and decrease the intracellular calcium contents. Which indicates that N-type calcium channel may play an important role in nociception of visceral inflammation induced by formalin.
