摘  要:目的观察不同浓度糖皮质激素对外周血CD4+T淋巴细胞凋亡的影响。方法取健康志愿者10名的全血样本,用RPMI1640培养液稀释一倍。分为五组,加入不同浓度氢化可的松:A组,0.3mg/L;B组,1.2mg/L,C组,36mg/L,D组,200mg/L,E组(对照组),0mg/L。以5%CO2培养箱培养4h,分离淋巴细胞,流式细胞术检测CD4+T淋巴细胞凋亡。结果0.3mg/L和1.2mg/L糖皮质激素致CD4+T淋巴细胞凋亡率分别为(8.74±1.3)%和(8.15±1.7)%,明显低于对照组的(10.84±2.1)%(P<0.05)。36mg/L和200mg/L糖皮质激素致淋巴细胞凋亡率分别为(11.59±2.2)%和(12.56±2.87)%,明显高于对照组(P<0.05)。结论低剂量糖皮质激素致T淋巴细胞凋亡率下降,而大剂量糖皮质激素则促淋巴细胞凋亡。Objective To investigate the influence of different doses of glucocorticoids on CD4^+ T lymphocyte apoptosis. Methods The blood samples of 10 healthy volunteers were diluted with RPMI 1640 one time, and then divited into 5 groups, in which hydrocortisone 0. 3 mg/L(group A), 1.2 mg/L(group B) ,36 mg/L(group C) ,200 mg/L(group D) or 0 mg/L(group E) was added. After cultured for four hours in 5%CO2 box, the lymphocytes were separated and CD4^+ T lymphocyte apoptosis was detected by flow cytometry. Results The lymphocyte apoptosis rate of group A and B was (8. 74 ±1.3)%and (8. 15±1.7)%, which was significantly lower than that of group E [(10.84 ±2. 1)% ] (P〈0.05), while that of group C and D was (11.59±2.2)% and (12.56±2.87) %, which was significantly higher than that of group E [(10.84+2.1)%] (P〈0. 05). Conclusion The lymphocyte apoptosis was depressed by lower dose of glucocorticoids,but promoted by higher dose of glucocorticoids.
