顺铂对去势雌性大鼠骨代谢影响的实验研究

摘　　要：目的通过对去势雌性大鼠的动物实验研究,观察顺铂对骨代谢、骨形态学的影响,探讨顺铂引起骨代谢变化的机理。方法3月龄雌性SD大鼠20只,随机分为对照组和化疗组,每组10只。各组大鼠均用1%戊巴比妥钠腹腔内麻醉行双侧卵巢切除术,术后1周开始用药,每两周用药1次,共6次。对照组腹腔内注射生理盐水1 ml;化疗组腹腔内注射顺铂2 mg/kg溶于1 ml生理盐水中。术后13周处死各组动物,处死前用代谢笼收集24 h空腹尿标本,自动生化分析仪测定尿钙浓度并计算出24 h排泌量,氯胺T氧化法测尿羟脯氨酸(HOP)浓度并计算出24 h排泌量;麻醉动物,开腹后下腔静脉取血离心,待放免法测骨钙素(BGP);取腰椎L3椎体,病理组织学切片。结果化疗组与对照组相比:前组骨组织形态学改变骨小梁数目减少、排列紊乱较后组严重。24 h尿HOP及尿Ca定量升高,骨吸收增强,差异有显著性,骨形成指标血BGP无差异。表明顺铂对骨有损害作用,与骨吸收增加有关,与骨形成无明显关系。结论顺铂对大鼠的骨代谢存在直接的不良影响,使骨吸收增加,表现为松质骨骨小梁数目减少、排列紊乱。Objective The objective of this cross-sectional study was to investigate the directive effect of DDP on bone metabolism. Methods Twenty female SD rats of three months were randomly divided into 2 groups, 10 rats a group, group Ⅰ ： blank operated; group Ⅱ ： operated and treated with DDP. Seven days after ovary resection all animals were treated. As control, animals in group Ⅰ received intrapefitioneally nomal saline; Animals in group Ⅱ were given intraperitioneally DDP 2 mg/kg/2 w, for there month. At the end of 13^th week the urinary sample was collected in 24 h for measurement of urinary calcium（Ca） and hydroxyproline（HOP）. The blood sample was collected for determination of osteocalcin （BGP）. Immediately after the animals were killed, by HE staining, the decalcified L3 vertebrae was made for histomorphologieal observation. Results In group Ⅱ , compared with group Ⅰ , the values of 24 h urinary Ca and 24 h urinary HOP remarkably increased; the values of BGP showed no significant difference between group Ⅰ and group Ⅱ ; In group Ⅱ the pathological changes of osteoporosis in L3 vertebra were more obvious than those in group Ⅱ , Conclusion Our present result showed that the negative influence of DDP on tissus nficrostrcture of lumbar vertebrae is due to an increase in bone resorption.
