摘  要:鉴定日本脑炎病毒(JEV)受体功能缺陷细胞3A10-3F与亲代细胞BHK-21膜分子表达的差异,以发现JEV受体候选分子。3A10-3F与BHK-21细胞膜蛋白用氯仿去脂法分别提取,通过二维电泳(2-DE)分离。经ImageMaster 2D软件分析比较后,选取差异点做液质联用质谱分析(LC-MS/MS)。发现有23个蛋白点在两组2-DE凝胶中有显著差异表达,LC-MS/MS鉴定了14个蛋白质,其中有4个膜蛋白,即钙结合蛋白annexin 1,annexin 2;电压依赖的离子通道(VDACs)蛋白VDAC1,VDAC2。上述结果为深入鉴定JEV受体分子特性与功能,阐明JEV致病机理提供了线索。To identify differentially expressed membrane proteins between 3A10-3F cells lacking the function of Japanese encephalitis virus (JEV) receptor and parental BHK-21 cells, hoping to provide clues to unveil the putative JEV receptors proteins, membrane-associated proteins from 3A10-3F and BHK-21 cells were obtained by the method of chloroform delipidation, and separated by two-dimensional electrophoresis (2-DE) respectively. The differential spots were analyzed with ImageMaster 2D software and identified by LC-MS/MS. There were twenty-three spots differentially expressed in 3A10-3F and BHK-21 cells gels. Fourteen proteins were identified by LC-MS/MS, and four of them were membrane proteins, which were annexin A1 and annexin A2, voltage-dependent anion channel (VDAC) 1 and 2. These data may provide important clues for identify the characteristics and function of JEV receptors and elucidate JEV pathogenesis.
