摘  要:目的研究藏花素诱导膀胱移行细胞癌T24细胞凋亡的作用及分子机制。方法通过MTT比色法分析藏花素对T24细胞增殖的影响;应用光镜观察细胞形态学的改变;Annexin V-FITC/PI荧光双标流式细胞术检测T24细胞周期动力学和凋亡诱导率的影响,RT-PCR和Western blot检测凋亡相关基因Bcl-2、Bax的表达变化。结果藏花素能明显抑制T24细胞的增殖,并可导致其形态学的改变;随藏花素作用时间的延长,G0/G1期细胞比例逐渐增加,S期和G2/M期细胞比例逐渐减少,早期凋亡细胞逐渐增多,同时可以上调Bax和下调Bcl-2的基因表达水平。结论藏花素可明显抑制T24细胞的生长,诱导肿瘤细胞凋亡,可能与Bcl-2和Bax等凋亡调控基因有关。Objective To investigate the apoptosis-induced mechanism of crocin on bladder transitional cell carcinoma cell line T24. Methods MTT assay was used to evaluate the inhibitory effect of crocin on the growth of T24 cells. Microscope was used to observe the morphological changes. The changes of cell cycle were measured by flow cytometry. Annexin V-FITC/PI double labeling flow cytometry was used to determine the cell apoptotic percentage. RT-PCR and Western blot were used to detect Bcl-2 and Bax expressions. Results The growth of T24 cells was inhibited remarkably in a dose-dependent and time-dependent manner. Distinct morphological changes of cell apoptosis were observed. Flow cytometric profiles revealed that crocin led to increase of the cells in G0/G1 phase and decrease of the cells in S phase, and the percentage of cell apoptosis was also increased. The mRNA and protein expressions of Bax increased and those of bcl-2 decreased. Conclusion Crocin exerts a fine inhibitory effect on the growth of human bladder transitional cell carcinoma cell line T24, maybe due to the apoptosis regulated by Bcl-2 and Bax pathway.
