中国南方汉族人触珠蛋白遗传多态性与OSAHS合并高血压的关系

摘　　要：目的探讨中国南方汉族人群触珠蛋白的遗传多态性对阻塞性睡眠呼吸暂停低通气综合征(OSAHS)合并高血压的影响。方法对103例OSAHS患者行多导睡眠监测,详细采集病史,测量其血压、身高、体质量和颈围,并计算体质量指数、Epworth嗜睡评分,测定血脂水平,根据是否合并高血压分为两组,抽提全血DNA和PCR扩增触珠蛋白基因的两个主要等位基因(Hp-1和Hp-2),用1%的琼脂糖电泳,分析触珠蛋白基因型Hp2-2、Hp2-1和Hp1-1,并与正常人群的触珠蛋白基因型进行比较。结果正常人群Hp基因不同表型构成为Hp2-2(59.1%)、Hp2-1(34.9%)和Hp1-1(6.0%);Hp-1基因频率为23.5%,男女分布无差异。高血压组与非高血压组相比,各表型所占百分比及Hp-1基因频率均无显著差异(P>0.05)。结论中国南方汉族人触珠蛋白遗传多态性可能与OSAHS合并高血压无关。Objective To investigate the effects of DNA polymorphism of the haptoglobin （Hp） gene on hypertension in patients with obstructive sleep apnea hypopnea syndrome （OSAHS） in Han population of south China. Methods A total of 103 cases of OSAHS underwent polysomnography amalysis. The medical history was collected, the blood pressure, height, body weight and neck circumference were measured, and the body mass index and Epworth sleepiness score were calculated. The patients were divided into two groups, hypertension group and non-hypertension group. DNA from the peripheral blood was extracted and two Hp alleles （Hp-1 and Hp-2） were determined by polymerase chain reaction （PCR）. Results The distributions of Hp genotypes in the normal population were Hp2-2 （59.1% ） , Hp2-1 （34.9%） and Hpl-1 （6.0%）. Hpl gene frequency was 23.5% , and there was no significant difference between the two genders. No significant difference was found between the hypertension group and non-hypertension group in the distributions of Hp genotypes and Hpl gene frequency （ P 〉 0.05）. Conclusion Hp polymorphism may not be associated with hypertension in patients with OSAHS in Han population of south China.
