摘 要:目的探讨双氢青蒿素对卵巢上皮性癌(卵巢癌)细胞体外增殖的影响,以及对卵巢癌细胞有丝分裂原激活的蛋白激酶(MAPK)磷酸化水平的影响。方法双氢青蒿素处理卵巢癌细胞株SKOV3和OVCAR3细胞后,四甲基偶氮唑蓝比色法检测SKOV3和OVCAR3细胞的体外增殖抑制效应,蛋白印迹法检测SKOV3和OVCAR3细胞中两种MAPK[即细胞外信号调节激酶(ERK)1/2和038蛋白激酶]的磷酸化水平,以未用双氢青蒿素处理的SKOV3和OVCAR3细胞作为对照。结果双氢青蒿素处理72h后,SKOV3和OVCAR3细胞的50%抑制浓度(Ic。)平均分别为(9.0±1.4)和(5.5±1.2)i^mol/L。双氢青蒿素处理后,SKOV3和OVCAR3细胞中ERK1/2的磷酸化水平明显降低,相对于对照细胞分别下降了64.2%和75.3%,差异均有统计学意义(P〈0.05);而p38蛋白激酶的磷酸化水平无明显变化,相对于对照细胞仅分别下降了8.5%和6.4%,差异均无统计学意义(P〉0.05)。结论双氢青蒿素能制卵巢癌细胞的体外增殖,该作用可能与其下调癌细胞内ERK1/2的磷酸化水平有关。Objective To determine the effect of dihydroartiminisin on the proliferation and phosphorylation of mitogen-activated protein kinase (MAPK) in SKOV3 and OVCAR3 ovarian cancer cell lines. Methods Methyl thiazolyl tetrazolium assay was performed to evaluate the anti-proliferative effect of dihydroartiminisin in SKOV3 and OVCAR3 cells, and Western blot was used to determine its effect on phosphorylation level of MAPK, including extra-cell regulated kinase (ERK) 1/2 and p38 protein kinase, in the two cell lines. Resalts Dihydroartiminisin inhibited the proliferation of ovarian cancer cells in vitro, with a mean of 50% inhibition concentration ( ICso ) at 72 h of (9.0 ± 1.4) Is, mol/L for SKOV3 and(5. 5 ± 1.2) μmol/L for OVCAR3 respectively. Compared to cells without dihydroartiminisin treatment, phesphorylation level of ERK 1/2 in SKOV3 and OVCAR3 cells treated with dihydroartiminisin decreased by 64. 2% and 751 3% respectively ( P 〈 O. 05 ), while phosphorylation of p38 protein kinase in SKOV3 and OVCAR3 only decreased by 8.5% and 6. 4% respectively (P 〉 0.05). Conclusion Dihydroartiminisin can inhibit the proliferation of ovarian cancer cell in vitro, probably through down-regulation of the phosphorylation of ERK 1/2 in ovarian cancer cells.
