摘 要:为选育高效产酸产氢细菌,采用1-甲基-3-硝基-1-亚硝基胍(NTG)、紫外线+亚硝酸(UV+亚硝酸)和5-溴尿嘧啶+紫外线(5-BU+UV)对产氢发酵细菌Ethanoligenens harbinense ZGX4进行诱变处理,显示出不同的诱变效应差异.NTG只得到一种类型产氢突变株;紫外线+亚硝酸法获得5种突变体类型,且出现一种自絮凝很强、产氢能力提高约50%的突变类型.5-溴尿嘧啶+紫外线法获得2种突变类型,约80%的突变株的释氢能力呈现负增长趋势.实验证明,紫外线+亚硝酸法的诱变效应最好,高产氢能力突变株显示其菌落直径增大、颜色透明的变化特征.In order to screen high efficient acid-producing and hydrogen-producing bacteria, the mutagenesis of 1-methyl-3-nitryl-1-nitrosoguanidine (NTG), ultraviolet rays (UV) and nitrous acid, 5-bromouracil and ultraviolet rays (5-BU + UV) on fermentative hydrogen-producing bacterium Ethanoligenens harbinense ZGX4 was studied, and different mutagenic effects were obtained. Only a kind of mutant was obtained by NTG mutagenesis; 5 kinds of mutants were attained by UV and nitrous acid, including a kind of H2-producing mutant with strong auto flocculation capacity and the improved H2-produicing ability of about 50% ; 2 kinds of mutants were obtained by 5-BU + UV and the H2-producing ability of about 80% mutants trended to decrease. Experimental results proved that the complex mutagenesis of UV and nitrous acid is the best way comparing with NTG and 5-BU + UV. The morphological characteristics of high efficient H2-producing mutants exhibit transparent color and bigger diameter.
