摘  要:用水溶性碳化二亚胺(EDC)法将新霉素(NEO)偶联于载体蛋白牛血清白蛋白(BSA)和鸡卵清蛋白(O-VA),合成免疫原BSA-NEO和包被原OVA-NEO,用红外扫描(IR)、SDS-PAGE法进行鉴定;用BSA-NEO免疫BALB/C小鼠,间接ELISA测定多抗上清(pAb)效价,阻断ELISA鉴定其敏感性,交叉反应试验鉴定其特异性。结果表明,BSA-NEO的红外光谱与BSA相比发生了改变,BSA-NEO的电泳条带与BSA的相比出现了明显的拖尾现象;通过系列ELISA鉴定,获得了高价、敏感、特异的pAb,为NEO残留免疫学检测方法的建立奠定了基础。Neomycin (NEO) was conjugated to carrier protein bovine serum albumin(BSA) and ovalbumin (OVA) by the method of carbodiimide (EDC),and immunizing antigen BSA-NEO,the coating antigen OVA-NEO were obtained in the same way.IR and SDS-PAGE were used to identify NEO artificial antigen.BALB/C mice were immunized with BSA-NEO,the titre of polyclonal antibody(pAb)was detected by indirect ELISA ,the sensitivity of pAb was identified by blocking ELISA and the specificity of pAb was identified by the method of cross-reaction test.The results showed that IR spectrogram of BSA-NEO has been changed when compared to BSA,the electrophoresis strap of BSA-NEO appeared the visible trail,when compared to BSA,the high-titer,sensitive and specific anti-NEO pAb have been produced in sera of immunized BALB/C mice,which made it possible to establish immunoassay of NEO residues in feed and animal food.
