摘  要:以牡丹(Paeonia suffruticosa)品种"金阁"的叶柄为外植体,研究不同基本培养基类型、激素种类及浓度配比在牡丹叶柄愈伤组织诱导与分化中的作用,进而优化牡丹愈伤组织的诱导与分化技术。结果表明:适宜牡丹"金阁"叶柄愈伤组织诱导的最佳培养基为:WPM+2,4-D 2.0 mg/L+6-BA1.0 mg/L+NAA1.0 mg/L+TDZ0.5 mg/L+CH300 mg/L;愈伤组织不定芽分化最佳激素组合:2,4-D 0.25 mg/L+TDZ 0.5 mg/L;继代周期以愈伤组织接种30 d为宜;不定芽在IBA浓度为2.0 mg/L时生根最好。The leafstalks of the cultivar "Jinge" of Paeonia suffruticosa were used as explants.The effects of the different basic medias,the different kinds and concentrations of hormones on callus induction and differentiation of leafstalks were studied,in order to optimized the techniques of on callus induction and differentiation of leafstalk.the results showed that the best media on induction of leafstalk callus was WPM+2,4-D2.0 mg/L+6-BA1.0 mg/L+NAA 1.0 mg/L+TDZ0.5 mg/L+CH300 mg/L.The best combination of hormone was 2,4-D 0.25 mg/L+TDZ 0.5 mg/L,The best rate of root induction from adventitious bud was 2.0 mg/L IBA.
