摘  要:目的揭示毒素清对老年肺炎导致小肠组织损伤的保护作用机制。方法复制克雷伯杆菌肺炎大鼠模型。将SD料龄大鼠随机分为对照组、模型组、毒素清组和左氧氟沙星组。观察肺及小肠组织的病理改变,测定组织含水量,采用免疫组织化学方法测定小肠组织白介素-1(IL-1)、肿瘤坏死因子-α(TNF-α)及巨噬细胞炎症蛋白-2(MIP-2)蛋白表达和原位杂交方法测定MIP-2mRNA的表达变化。结果模型组大鼠的肺脏及小肠组织损伤明显,肺脏和小肠组织含水量增高;模型组小肠组织IL-1、TNF-α及MIP-2的表达和MIP-2mRNA的表达均较对照组显著增强(P<0.05);与模型组比较,毒素清组和左氧氟沙星组肺脏及小肠组织病理损伤明显改善,组织含水量降低,小肠组织IL-1、TNF-α及MIP-2蛋白表达和MIP-2mRNA的表达减弱(P<0.05)。结论IL-1、TNF-α及MIP-2的表达上调参与了小肠组织损伤,毒素清通过下调其表达来保护小肠组织。Objective To study the mechanism of protective effect of DuSuQing on small intestine injury induced by pneumonia in the aged. Methods The model of rats with Klebsiella pneumonia was established. The aged SD rats were randomly divided into control, model, DuSuQing and levofloxacin groups. The pathological changes and water content of small intestine and lung were observed and determined. IL-I, TNF-α and MIP-2 protein expressions were measured by immunohistochemistry. MIP-2mRNA expression was detected by in situ hybridization. Results The rats of model group had obvious intestine injury, the increased water content, but those of DuSuQing and levofloxacin groups had less injury and the decreased water content. The expressions of IL-1, TNF-α, MIP-2 and MIP-2mRNA of model group were significantly higher than those in the control group, while those of DuSuQing and levofloxacin groups were significantly lower than those of model group. Conclusions The intestine injury induced by Klebsiella pneumonia is related to IL-1, TNF-α, MIP-2 and MIP- 2mRNA. To decrease the excessive expression of IL-1, TNF-α, MIP-2 and MIP-2mRNA might be the main mechanism of protective effect of DuSuQing on intestine injury.
