摘 要:目的研究中国甘肃汉族、回族人群E-选择素基因第2外显子G98T和第4外显子S128R多态性与冠心病的相关性,以及对血清E-选择素水平的影响。方法采用聚合酶链反应-限制性片断长度多态性(PCR-RFLP)技术,分别检测甘肃汉族、回族冠心病患者108例、42例及两民族健康对照者96例、50例。检测他们的E-选择素基因多态性,同时采用酶联免疫吸附实验(ELISA)检测冠心病患者和对照者血清E-选择素水平。结果E-选择素基因S128R基因频率和等位基因频率在两民族冠心病组和对照组比较差异有显著性(P<0.05)。SR基因型携带者的E-选择素血清水平明显高于SS基因型者(40.26±3.61vs22.21±4.23)(P<0.01)。E-选择素G98T各基因型分布在两民族冠心病组和对照组之间比较差异无显著性(P>0.05)。结论E-选择素S128R基因多态性与冠心病的发病有关联,并可影响血清E-选择素的水平,R等位基因可能是冠心病的候选基因,E-选择素基因G98T多态性在冠心病的发病中可能不起直接重要作用。Objective To study the relationship between the polymorphism of the second and fourth exon G98T and S128R of E-seleetin and coronary heart disease (CHD) and its effect on E-selectin level of Han and Muslim nationality. Methods The polymorphism and level of E-seleetin of 108 CHD patients and 42 healthy people of Han and Muslim nationality in Gansu were determined by PCR-RFLP and ELISA respectively. Results There was significant difference in the frequencies of S128R and allele between CHD and control group ( P 〈 0.05 ). E-selectin level of SR genotype carriers (40. 26 -+ 3.61 ) was significantly higher than that of those with SS ( 22. 21 -+ 4. 23 ) ( P 〈 0. 01 ). There was no significant difference in G98T genotype distribution between CHD and control group ( P 〉 0. 05 ). Conclusions S128R gene polymorphism of E-selectin might be related to CHD, could influence E-selectin level. R allele might be candidate gene of CHD. G98T polymorphism of E-seleetin could not play directly important role in CHD.
