单克隆抗体F36/22分离和检测小鼠移植性乳腺肿瘤抗原的研究

摘　　要：作者应用免疫印迹法检测到单克隆抗体F36/22能识别小鼠移植性乳腺癌抗原.用SepharoseCL-4BA单克隆抗体F36/22免疫亲和层析法分离纯化了BALB/c小鼠移植性乳腺肿瘤抗原,固相放射免疫法检测纯化188.4倍,SDS-PAGE及免疫印迹检测其分子量为60kd。A transplantable murine breast tumorantigen (called JC tumor) has been recog-nized by the detection of McAb F36/22 withthe method of immunoblotting. JC tumorantigen was isolated by Sepharose-CL-4B,then was purified by McAb F36/22 affinitychromatography. Solid phase RIA detectiondemonstrated that the specific activity ofthe antigen which was purified by 188.4times was 148000 cpm/mg protein. SDS-PAGE and immunoblot analysis revealedthat the tumor antigen possessed a molecularweigh of 60kd. Competitive binding RIAindicated that JC tumor antigen inhibitedthe combining of ductal carcinoma antigenwith McAb F36/22. It implied that somegroups (antigen determinant)on JC tumorantigen were the same as or similar tothose on ductal cancer antigen.
