摘 要:目的研究大黄治疗慢传输型便秘大鼠结肠肌间神经丛内胆碱能(AchE)神经的变化,从神经病理学角度初步探讨大黄治疗慢传输型便秘的机制。方法建立大鼠慢传输型便秘模型,大黄治疗后利用铺片技术制作结肠肌间神经丛标本,采用组织化学技术行结肠肌间神经丛内AchE神经染色。结果结肠肌间神经丛铺片组化染色发现便秘大鼠结肠肌间神经丛内胆碱能神经分布较稀疏,数量较少,但胞体增大,染色加深。大黄治疗组大鼠结肠肌间神经丛中的胆碱能神经分布趋于正常,胆碱能阳性神经元较多(阳性细胞数25.71±5.36与便秘组14.32±4.60,P<0.01;核浆比例0.71±0.21与便秘组0.58±0.19,P<0.01)。结论慢传输型便秘大鼠结肠传输功能障碍与肠壁内胆碱能的病理改变和(或)功能障碍有关。大黄使便秘大鼠结肠肌间神经丛中的胆碱能神经分布趋于正常,可能是其调节结肠功能、治疗慢传输型便秘的机制之一。Objective To study the effect of rhubarb on the changes in cholinergic nerves in colonic myenteric plexus for the chronic slow transit constipation rat. Methods Slow transit constipation model of rat was reproduced by feeding diphenoxylate. The changes in chotinergic nerves in colonic myenteric plexus were studied by histochemical technology. Results In the colonic myenteric plexuses of rats with slow transit constipation,cholinergic nerves and cholinergic cells were significantly decreased with denser immunostainning,but its volume increased. After treated by rhubarb,cholinergic nerves and cholinergic cells tended to be normal (positive cells were 25.71 ± 5.36 vs STC model group 14.32 ± 4.60, P〈0.01;the ratio of nuclei/cytoplasm was 0.71± 0. 218 vs STC model group 0. 58±0.19 ,P〈0.01). Conclusion Dysfunction of colon transit is related with the changes in cholinergic nerves of colonic myenteric nerve plexus in rats with slow transit constipationl. Rhubarb can adjust the cbolinergic nerve supply in STC rats,which maybe a mechanism of treating STC.
