摘 要:目的探讨钙通道拮抗药非洛地平是否对造影剂诱导的肾小管上皮细胞损伤具有保护作用及其机制。方法培养大鼠肾小管上皮细胞(NRK52E)与非洛地平(10-5mol.L-1)作用12 h后,加入造影剂(110 mgI.L-1)作用0.5,1.0 h,采用乳酸脱氢酶(LDH)检测细胞损伤,倒置显微镜观察细胞形态变化,流式细胞仪检测细胞凋亡,共聚焦显微镜测定细胞内钙和反应氧产物水平。相同渗透压甘露醇作对照。结果泛影葡胺作用0.5,1.0 h明显诱导细胞损伤或细胞凋亡,造影剂组细胞内钙和反应氧产物随时间进行性增加,显著高于相同渗透压甘露醇组和对照组;非洛地平降低细胞内钙和反应氧产物水平,同时降低细胞损伤和细胞凋亡。结论非洛地平通过降低细胞内钙超载和氧化应激对造影剂诱导的肾小管上皮细胞损伤发挥保护作用。Objective To investigate whether felodipine plays renoprotective effect on tubular epithelial cell injury induced by contrast media. Methods NRK-52E cells were treated with concentrations ( 10.5 mol · L^-1 ) of felodipine 12h before contrast media was added. At 0.5, 1.0 h with contrast media ( 110 mgI · L^-1 ), cell injury was assessed by LDH, cell morphologic change and cell apoptosis were evaluated with inverted microscope and flow cytometry respectively; Intracellular Ca^2+ and reactive oxygen species C ROS) were analyzed using eonfocal microscope. Mannitol with the same osmolality was used as control. Results Incubation with contrast media significantly induced tubular cell damage or apoptosis cells at 0.5 h and 1 h, meanwhile, intracellular Ca^2+ and ROS were increased progressively in CM group at 30 min and 60 min compared with mannitol and control groups. Flodipine decreased cell injury and apoptosis, as well as intracellular Ca^2+ and ROS. Conclusion Felodipine plays a renoprotective effect on the contrast media-induced renal tubular cyotoxicity by decreasing intracellular Ca^2+ overload and oxidative stress.
