摘 要:[目的]探讨环氧化酶2抑制剂尼美舒利对乳腺癌细胞株MCF-7的放射增敏作用并初步探讨其机制。[方法]克隆形成实验检测尼美舒利对MCF-7的放射增敏作用;电镜观察肿瘤细胞形态学改变;流式细胞仪(FCM)检测肿瘤细胞调亡率及Bcl-2、Bax蛋白的表达。[结果]不同浓度的尼美舒利对乳腺癌细胞MCF-7均有放射增敏作用,且呈剂量依赖性关系;尼美舒利与射线照射联用能够上调Bax的蛋白表达,同时抑制Bcl-2蛋白表达,明显增加细胞凋亡率。[结论]COX2抑制剂尼美舒利对人乳腺癌MCF-7细胞株具有明显的放疗增敏作用。[Objective] To investigate the effect of radiosensitizing effect of COX-2 inhibitor -Nimesulide on breast cancer cell line (MCF-7) in vitro and try to research the possible radiosensitizing mechanisms: [ Methods] The clonogenic assay was performed to determine the radiosensitizing effect of Nimesulide on MCF-7 cell line. The change of morphological feature in MCF-7 was observed by electron microscopy. The apoptosis and the expression in protein of Bcl-2 and Bax were measured by flow cytometry (FCM) . [Results] The different dose of Nimesulide could significantly enhance the MCF-7 cells radiosensitivity effect in a dose-dependent manner. The expression of the protein of Bax was up-regulated, the expression of the protein of Bcl-2 was inhibited and the apeptosis was increased. [Gonclusion] It is suggested that Cox-2 inhibitor obviously enhance the radiosentivity effect of human breast cancer cell line MCF-7.
