7-硝基吲唑对深低温停循环大鼠的脑保护作用

摘　　要：目的:探讨神经元型一氧化氮合酶(nNOS)在深低温停循环(deep hypothermic circulatory arrest,DHCA)大鼠中的脑损伤作用。方法:154只健康SD大鼠随机分为4组:假手术组、DHCA组、7-硝基吲唑(7-NI)组和花生油组,后3组再分为DHCA术后2,6,12和24 h 4个亚组。假手术组与24 h亚组各16只大鼠,其余亚组各10只大鼠,冰块包埋体温降至21℃时阻断双侧颈总动脉建立DHCA模型。观察特异性nNOS抑制剂7-NI对脑水肿,脑组织光镜、电镜结构和脑组织丙二醛含量的影响。结果:与DHCA组相比,7-NI能减轻脑水肿(P<0.05),改善脑细胞形态,并能降低丙二醛含量(P<0.05)。结论:nNOS在DHCA脑损伤中起细胞毒性作用,7-NI有希望延长DHCA安全时限。Objective： To investigate the function of neuronal nitric oxide synthase （nNOS） in deep hypothermic circulatory arrest. Methods： One hundred and fifty-four SD rats were randomly divided into 4 groups ： fake operation group （ n = 16 ）, DHCA group （ n = 46 ）, 7-nitroindinazole （7-NI） group （ n = 46 ）, arachisoil group （ n = 46 ）. The last 3 groups were each divided into 4 subgroups according to the time phase after DHCA, which was 2 h（n =10）, 6 h（n =10）, 12 h（n =10）, 24 h（n =16）. Imbedding rats by ice cubes to reduce the temperature and occlusion both common carotid arteries while the temperature reach 21℃. The effect of nNOS inhibitor 7-NI on ischemia edema, structure and contents of MDA in cerebral were studied. Results： 7-NI significantly lessened ischemic edema（P 〈0.05 ）, improved neuronal cell degeneration, decreased MDA content of the brain. （P 〈 0.05 ）. Conclusion： nNOS may act as a neurotoxic mediator in DHCA, and 7-NI may be used to prolong time limited of DHCA.
