摘 要:目的:从脑细胞周期的角度揭示熟地黄防治脑衰老的作用机制。方法:36只SD大鼠随机分为正常组、模型组、天保宁组和熟地黄组,采用D-半乳糖造模同时服用相应药物45d。在检测大鼠行为学、脑组织生化指标、细胞衰老特异代谢产物SA-β-gal阳性表达细胞数的基础上,采用流式细胞术检测细胞周期、免疫组化法检测海马CA1区P53,P21,P16的表达。结果:熟地黄能一定程度促进细胞周期从G0/G1期向S期进展,抑制海马CA1区CDK抑制基因蛋白P53,P21,P16的表达。结论:熟地黄能调控细胞周期相关基因P53,P21,P16表达。Objective: To study the mechanism of prepared Radix Rehmanniae (ShuDi) on brain ageing based on cell cycles. Method: Thirty-six Sprague-Dawley rats were divided into 4 groups randomly, that were normal, model, Tianbaoning and prepared Radix Rehmanniae (ShuDi) group. Besides prophase study on rats inteUigence,biochemical changes of brain tissue and special products SA-β-gal of brain cells,cell cycles were further determined by flow cytometry and P53, P21 and P16 express in CA1 hippocampus zone were also observed by immunohistochemistry. Result: Prepared Radix Rehmanniae (ShuDi) could prompt the progression of cell cycles in some extent,decrease the expression of P53, P21 and P16 in CA1 hippocampus zone were decreased. Conclusion: Prepared Radix Rehmanniae (ShuDi) could regulate cell cycle related gene P53, P21 and P16 expression in CA1 hippoeampus zone.
