摘 要:目的:观察低氧训练对大鼠肾组织细胞凋亡及HIF-1α、bax、bcl-2表达的影响,为探讨低氧训练适应机理提供依据。方法:60只SD大鼠按体重随机分为6组,每组10只,即:常氧组(C)、高住8h组(8hHi)、高住12h组(12hHi)、常氧运动组(T)、高住低练8h组(8hHiLo)和高住低练12h组(12hHiLo)。T、8hHiLo、12hHiLo组大鼠每天在坡度为0的动物跑台上以25m/min的速度训练1h。训练后,将8hHi、8hHiLo组和12hHi、12hHiLo组分别放入氧浓度为12.5%(相当于海拔4000m)的低氧舱内8h和12h,5d/周。实验周期为4周。最后一次实验结束后24小时取材,采用HE染色、原位末端脱氧核糖核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL)及蛋白免疫组织化学法检测各组大鼠肾组织细胞凋亡和HIF-1α、bcl-2、bax表达,分析各指标之间的相关关系。结果:(1)与常氧组相比,高住12小时组、常氧运动组及高住低练组凋亡指数均显著增加(P<0.05);与常氧运动组比较,12小时高住低练组凋亡指数增加,具有统计学意义(P<0.05);8小时高住低练组凋亡指数比8小时高住组显著增加(P<0.05),12小时高住低练组凋亡指数比12小时高住组显著增加(P<0.05);与8小时高住低练组相比,12小时高住低练组凋亡指数显著增加(P<0.05)。(2)高住组、常氧运动组及高住低练组与常氧组相比HIF-1α、bcl-2、bax、bax/bcl-2均具有显著性差异(P<0.05);8小时和12小时高住低练组与常氧运动组相比,bcl-2、bax、bax/bcl-2亦具有显著性差异(P<0.05);12小时高住低练组bcl-2、bax、bax/bcl-2比12小时高住组显著增加(P<0.05),与8小时高住低练组相比,12小时高住低练组bcl-2、bax、bax/bcl-2显著增加(P<0.05);(3)凋亡指数与bax及bax/bcl-2、HIF-1α与bax呈正相关(P<0.05)。结论:(1)低氧训练可诱导大鼠肾组织HIF-1α、bcl-2以及bax蛋白表达,细胞凋亡指数及病理损伤与运动时低氧刺激有关,以高住低练12小时组最明显。(2)bcl-2与bax参与调控肾�Objective To study the effect of simulated altitude training on the expression of HIF-1α, bax, bcl- 2 and the renal apoptosis. Methods Sixty male Sprague-Dawley rats were randomly assigned into two groups: control group (Ⅰ) and training group ( Ⅱ ) (n= 10). Group I was divided into normoxic group (C) and living at high altitude for 8h (8hHi) and 12h (12hHi) groups, and group 11 was divided into normoxic training (T) and training at high altitude for 8h (8hHiLo) and for 12h (8hHiLo) groups. Hypoxic chamber with 12.5% concentrations of oxygen was used as simulation of 4000m altitude. The rats in group Ⅱ underwent treadmill running at the speed of 25m/min for lh without incline per day and then were sent into the hypoxic chamber for 8h and 12h respectively every day for 4 weeks. The expression of HIF-α, bax, bcl-2, TUNEL was determined. Results (1) Increase in the TUNEL dyeing slice in groups 12hHi, T, 8hHiLo and 12hHiLo wassignificant than in group C (P〈0.05), and was significant between group 8hHiLo and group 12hHiLo (P〈0.05). Increase in the TUNEL in groups 8hHiLo and 12hHiLo was significant than in groups 8hHi and 12hHi (P〈0.05). The expression in group 12hHiLo was even more significant than in group T (P%0.05). (2) The expression of HIF-α, bcl-2, bax, bax/bcl-2 in groups 8hHi, 12hHi, T, 8hHiLo and 12hHiLo increased more significantly than in group C (P〈0.05). There was statistical significance between group 8hHiLo and group 12hHiLo (P〈0.05). The expression of HIF-1α, bcl-2, bax and bax/bcl-2 in group 12hHiLo increased more significantly than in group 12hHi (P〈0.05). As compared with the group T, the expression was even more significant in groups 8hHi and 12hHiLo (P〈0.05). (3) Apoptotic index was highly related to the bax and bax/bcl-2; and HIF-1α was highly related to bax.
