草木樨药材香豆素的含量测定

摘　　要：目的:探讨草木樨药材中香豆素提取与含量测定方法,从而为草木樨的质量控制奠定相应基础。方法:采用HPLC测定方法,以香豆素含量为评价指标从提取溶剂、药材粒径、提取时间、提取次数、提取方法等多个方面进行超声、索式、回流3种方法进行了提取方法研究。结果:采用超声提取方法较好,提取条件是60～80目药材0.25g,加入50mL50%甲醇超声提取30min,其提取效果均好于其他几种方法,其平均加样回收为99.49%,RSD%为0.53%。结论:该提取方法比较简单,对香豆素的提取效果较好,基本上反映出草木樨药材中香豆素的含量,可以作为草木樨香豆素含量测定的评价方法。OBJECtIVE To study on extraction process and determination method of coumarin from Melilotus suaveolens Ledeb. METHODS HPLC with UV detector was used to analyze the extraction from Melilotus suaveolens Ledeb, coumarin was used as standard substances and index,main factors influencing the extraction of coumarin, such as extraction solvent, ex- traction times, extraction time, extraction method and different herb powder were studied. Optimized conditions for the extrac- tion were obtained. RESULTS The selected method was as follows： ultrasonic with methanol for 30 min： 0. 25 g of 60-80 mesh herb powder, added with 50 mL of 50% methanol, the mean recovery and RSD were 99. 49% with RSD of 0. 53%. CONCLUSION The extraction method is simple and effective, the determination method could be used for evaluate the content of eoumarin in Melilotus suaveolens Ledeb.
