摘 要:目的评价异氟醚注射液是否对大鼠各脏器、系统产生损害及损害程度和可逆程度。方法 100只 SD 大鼠随机分为生理盐水组(生理盐水3.0 ml/kg),溶媒对照组(30%脂肪乳3.0ml/kg),异氟醚注射液高剂量组(1.1 ml/kg),中剂量组(0.9 ml/kg)和低剂量组(0.7 ml/kg),每组20只.雌雄各半。每天以等容量(3.0 ml/kg)、等速度(1.5 ml/kg·10 s)静脉给药一次.连续给予30 d。期间记录动物一般情况;在给药期结束(最后一次给药后24 h)、恢复期结束(最后一次给药后两周)两时点每组各选择一半动物采血进行血液学及生化学指标检测,处死后取所有脏器及给药部组织标本行病理学检查。结果给药前、给药期结束、恢复期结束三个时点各组间体重差异无统计学意义。恢复期结束高剂量组红细胞均数的95%可信区间位于正常参考值范围内。恢复期结束五组血清尿素浓度均数的95%可信区间均未超出正常参考值范围。给药期结束和恢复期结束未发现大鼠器官特异性组织病理学损害。结论长期静脉注射异氟醚注射液未对大鼠各脏器和系统产生明显毒性损害。Objective To estimate whether emulsified isoflurane induces impairments of rat's organs or systems and whether the impairments are reversible. Methods One hundred Sprague-Dawley rats were randomly divided into five groups (10 male rats, 10 female rats for each group): control group one(with physiologic saline solution 3.0 ml/kg), control group two(with 30% fat emulsion 3.0 ml/kg), high-dose group (with emulsified isoflurane 1.1 ml/kg), medium-dose group (with emulsified isoflurane 0.9 ml/kg) and low-dose group (with emulsified isoflurane 0.7 ml/kg). All rats were daily given corresponding drugs which diluted into the same volume (3.0 ml/kg) with 30% fat emulsion via tail vein approach at the same rate (1.5 ml/kg · 10 s) for thirty consecutive days. At the end of twenty-four hours and two weeks after the last injection, half of the rats in each group were killed for the determination of hematology and biochemistry values, and tissue specimens were collected for histopathologic investigation. Results Red blood cell counts of high-dose emulsified isoflurane group was lower than that of other groups at the end of two weeks after the last injection (P〈0.05), but its 95% confidence interval was in the normal range. The 95% confidence intervals for mean values of serum urea in all five groups at the end of two weeks after the last injection were in the normal range. Histopathologic investigation showed that no specific lesions were found in any organ or tissue in all rats. Conclusion Long-term intravenous administration of emulsified isoflurane does not impair rat's organs and systems.
