摘 要:目的观察丙泊酚不同剂量、不同持续输注时间对老年大鼠空间学习记忆的影响。方法 18月龄雄性 SD 大鼠56只,随机均分为七组,按照丙泊酚两种剂量[小剂量组(S)30 mg·kg^(-1)·h^(-1),大剂量组(L)50 mg·kg^(-1)·h^(-1)]、不同输注时间(2,4 h)和饲养天数(1,7 d)分为:S12组、S14组、L12组,S72组、S74组和 L72组,另设一对照组(C 组)。除对照组外,各组大鼠经尾静脉恒速输注不同剂量和持续输注时间的丙泊酚。分别在给药后1和7 d,对大鼠行持续5 d 的 Morris 水迷宫测试。记录大鼠每日寻找固定平台的时间(潜伏期)和有效搜索策略所占百分比。结果水迷宫测试的第3天和第5天,S12组的潜伏期(49.8±14.0)s 和 L12组的潜伏期(49.0±9.1)s 明显长于 C 组的潜伏期(21.7±7.2)s(P<0.05);L12组的游泳行程(530.6±157.4)cm 明显长于 C 组的游泳行程(21 8.5±49.8)cm(P<0.05);S12组的正常认知策略(53.1±10.0)%和 L12组的正常认知策略(56.3±10.3)%明显小于 C 组的正常认知策略(87.5±9.5)%(P<0.05)。结论丙泊酚静脉注射后1d 可致老年大鼠空间认知能力下降,但对给药后7 d 认知能力无显著影响。Objective To investigate the effect of propofol on spatial cognitive dysfunction in aged rats. Methods Fifty-six 18 months old male SD rats weighing (549.2±16.4) g were randomly equally divided into 7 groups according to propofol infusion dose [small dose group (S) 30 mg · kg^-1 · h^-1 ,large dose group (L) 50 mg · kg^-1 · h^-1) ,infusion time and raised date after propofol infusion [small dose propofol 2 h/d-ld group (S12 group),small dose propofol 4 h/d-1d group (S14 group), large dose propofol 2 h/d-1d group (L12 group), small dose propofol 2 h/d-7d group (S72 group), small dose propofol 4 h/d-7d group (S74 group) ,large dose propofol 2 h/d-7d group (1.72 group) and control group (C group). Excluded control group,rats in the other six groups were anesthetized with propofol intravenously through the tail vein. The induction dose of propofol for six groups was 10 mg/kg and anesthesia maintenance dose was 30 mg · kg^-1 · h^-1 for small dose group and 50 mg· kg^-1 · h^-1 for large dose group. The infusion time of propofol was 2 h or 4 h according to group division. The rats were kept sleeping during propofol infusion. When the propofol infusion was stopped and rats were expected to wake and keep alive for l d or 7 d. All of the rats underwent Morris water-maze (MWM) test four times a day for 5 consecutive days from the 1^st or 7^th day after propofot intravenous anesthesia. The latency period,swimming speed and normal strategy of MWM test were recorded. Results In 1 d groups,on the 3ra and 5^th day of MWM test,the latency period of S12 group and 1,12 group was significantly longer than that of the control group (P〈0.05), and the normal strategy of S12 group and L12 group was significantly lower than that of the control group (P〈 0.05). Conclusion Propofol continuous intravenous infusion may cause transient spatial cognitive dysfunction in aged rats,but does not have long term effect.
