摘 要:以加拿大披碱草(Elymus canadensis L.)与披碱草(E.dahuricus Turcz.)、圆柱披碱草(E.cylindricus(Franch.)Honda)2个种间杂种F1及其亲本为试材,利用AFLP分子标记技术,对2个杂种F1及其亲本的遗传差异性进行比较分析,以期为杂种鉴定及后代选育提供分子依据。结果表明:用筛选出的13个适宜引物共扩增出1247个AFLP位点,其中多态性位点1042个,多态性位点比率高达83.56%;各供试材料的遗传距离(GD)变动在0.4276~0.7486之间,以GD值0.65为基准,5个材料聚为2类,第1类为加拿大披碱草、圆柱披碱草和加拿大披碱草×圆柱披碱草杂种F1,第2类为披碱草、加拿大披碱草×披碱草杂种F1。In order to provide a molecular basis for identification of hybrids and breeding of progenies, the genetic difference of Elymus canadensis L. × E. dahuricus Turcz. hybrid F1 , E. canadensis × E. cylindricus (Franch.) Honda hybrid F1 and their parents were compared and analyzed by AFLP molecular marker technique. The results show that 1247 loci of AFLP markers were amplified with 13 suitable primer combinations,and 1042 loci among which were polymorphic and the percentage of polymorphism loci reached to 83.56%. The genetic distance of test plants was ranged from 0. 4276 to 0. 7486. Taking the genetic distance value of 0.65 as threshold,5 test plants were clustered into 2 groups:the first group included E. canadensis,E, cylindricus,and E. canadensis × E. cylindricus hybrid F1 ;the second groups included E. dahuricus and E. canadensis × E. dahuricus hybrid F1.
