摘 要:为了弄清尿毒症状态或环境是否激活外周血单个核细胞诱导IL-1β、TNF-α与IL-6基因表达。方法 选择了7名规律性铜仿膜血透患者、11名尿毒症非透析患者及10名正常对照者,采用反转录多聚酶链反应(RT-PCR)与细胞原位杂交技术对外周血单个核细胞中IL-1β、TNF-α、IL-6的基因表达进行观察。结果 正常对照者外周血单个核细胞中IL-1β、TNF-α、IL-6的基因表达检测不出,尿毒症非透析患者与血透患者均有IL-1β、TNF-α、IL-6的基因表达,但尿毒症非透析患者的基因表达水平与血透患者相比明显的较低。结论 尿毒症状态或环境或其相关因素可激活外周血单个核细胞,这种预激活状态可能与尿毒症患者机体抵抗力下降有关。To determine whether uremia per se can activate peripheral blood mononuclear cells (PBMCs). Methods Maintained hemodialysis patients with cuprophane, non-dialysis uremic patients and healthy volunteers were selected to investigate IL-1β、 TNF-α and 11,6 mRNA in PBMCs by RT-PCR and cells in situ hybridization. Results IL-1β,TNF-α and IL-6 mRNA in PBMCs were undetected in healthy volunteers, but these cytokine gene expression were detected in hemodialysis patients and non-dialysis uremic patients. IL-1β、TNF-α and IL-6 mRNA level was lower in non-dialysis uremic patients than that in hemodialysis patients. Conclusion Uremia per se or its related factors may activated PBMCs and the preactivation of PBMCs might be associated with the body resistance against infection in uremic patients.
