新疆艾比湖细菌视蛋白基因资源研究

摘　　要：针对新疆艾比湖这一特定生境中的嗜盐古菌的细菌视蛋白(bacterio-opsin,bop)基因资源进行了研究.通过PCR方法扩增并测定了11株菌株的编码细菌视蛋白螺旋C至螺旋G片段的基因序列.根据该片段基因序列进行的系统发育分析表明,艾比湖获得的bop基因大部分聚类到同一个类群,具有明显的地域特征.将翻译出的氨基酸序列与已报道的相应片段进行对比表明,从艾比湖菌株获得的序列与其他菌株来源的序列间存在明显差异.这些天然的突变体,尤其是菌株AB9、AB27、AB47,都是研究细菌视紫红质(bacteriorhodopsin,BR)蛋白结构和功能的良好材料,而细菌视紫红质是人工视网膜和光子开关等的良好生物材料.The resource of bacterio-opsin （bop） gene from the halophilie archaea inhabiting in Aibi lake in Xinjiang region was researched. The gene fragment encoding the bop segment of helix C to helix G was obtained by polymerase chain reaction （PCR）respectively from eleven isolates, and the sequences were determined subsequently. According to the phylogenetic analysis based on the gene fragments, it was considered that the bop genes from Aibi lake formed a microbiological group, which showed its own regional trait. On the other hand, similarity alignment between the deduced amino acid sequences from Aibi lake isolates and other reported strains showed that the former was obviously different. These natural bop gene mutants, especially from strain AB9, AB27 and AB47, are good materials for the structural and functional research of the bacteriorhodopsin （BR） protein which is the best biomaterial for man-made retina and photon switch.
