缬沙坦对糖尿病大鼠肾脏的保护作用及其机制

摘  要:目的探讨血管紧张素受体拮抗剂缬沙坦对糖尿病大鼠肾脏的保护作用及其机制。方法36只雄性SD大鼠随机分成正常组、糖尿病组和缬沙坦治疗组。实验2、4、8、12周后分别处死大鼠。光学显微镜下观察肾组织病理改变,逆转录-聚合酶链反应测定肾脏结缔组织生长因子(CTGF)mRNA表达,免疫组织化学测定肾脏CTGF蛋白的表达。结果与正常组比较,糖尿病组和缬沙坦治疗组大鼠的血糖、血尿素氮、血肌酐、尿白蛋白排泄率均显著增高(P值分别<0.01、0.05),肾脏CTGF mRNA和CTGF蛋白表达显著增加(P值分别<0.01、0.05)。缬沙坦治疗组大鼠血尿素氮、血肌酐、尿白蛋白排泄率较糖尿病组大鼠均有不同程度的显著降低(P值分别<0.01、0.05),肾脏CTGF mRNA和CTGF蛋白表达显著降低(P值均<0.05)。结论缬沙坦对糖尿病大鼠肾脏保护的作用可能与减少肾脏CTGF mRNA和CTGF蛋白的表达有关。Objective To investigate the protective effect of Valsartan, an antagonist of angiotensin receptor, on diabetes mellitus in rats and to study the possible mechanisms. Methods Thirty-six male SD rats were randomly divided into normal control group, diabetes mellitus (DM)group and diabetes mellitus plus Valsartan group (ARB group). The rats were sacrificed at 2, 4, 8 and 12 weeks of the experiment. The renal morphology was observed under light microscopy; the mRNA expression of connective tissue growth factor (CTGF) in kidney was determined by reverse transcription-polymerase chain reaction (RT-PCR); and the expression of CTGF protein in the renal tissues was examined by immunohistochemistry. Results The serum glucose, blood urea nitrogen (BUN), creatinine (Cr), urine albumin excretion rate (UAER) and expression of CTGF mRNA and protein in the DM group and ARB group were significantly higher than those in the control group (P〈0.01 or 0.05). All the above indices in ARB group were significantly lower than those in the DM group (P〈0.01 or 0.05). Conclusion The protective effect of Valsartan on diabetes mellitus may be related to the decrease of renal expression of CTGF mRNA and protein.
