电针对大鼠脑缺血急性期治疗机理及神经保护作用的研究

摘　　要：目的探讨电针对大鼠脑缺血急性期治疗机理及脑组织自由基水平与神经功能缺损的相关性。方法53只健康的雄性Wister大鼠,随机分为针灸组、模型组、对照组。采用光化学法复制局灶性脑缺血急性期动物模型。分别在术后3h、24h检测脑组织中MDA含量和SOD活性,同时进行神经行为学观察和神经功能缺损评分。结果模型组脑缺血后3 h MDA含量已明显增高,24 h其值为正常组的3倍(P<0.01)。电针组24 h MDA含量相对降低,由(4.02±0.31)mol.ml-1降为(2.01±0.54)mol.ml-1,与模型组相比,P<0.05;电针组24h SOD的活性明显增高,由(66±1.24)nu/mg pro上升为(103±2.54)nu/mg pro,与模型组相比,P<0.05。神经功能缺损ZeaLonga法评定显示,24 h电针组的神经功能缺损程度明显低于模型组,其神经功能缺损程度与脑组织MDA含量呈负相关,与SOD活性呈正相关。结论电针可以通过干预大鼠缺血性脑组织自由基水平,对脑组织起保护作用,同时自由基的水平与脑缺血后神经功能的缺损程度有密切关系(P<0.05)。*Objective To study the mechanism of electric acupuncture on acute cerebral ischemia and free radical level in brain tissue correlated with degree of neurologic impairment in rats. Methods 53 Wister rats were divided into electric acupuncture therapy group and control group. Rats＇focal cerebral ischemia in acute stage was induced by photochemistry method. Changes in contents of MDA and in activity of SOD in brain homogenate were observed at 3h and 24h. The degree of neurologic impairment was examined by Zea Longa＇law. Results After 3h the contents of MDA in brain ischemia tissue increased, and at 24h it increased by 3 times compared with control group. MDA contents at 24h in the acupuncture therapy group dropped significantly from 4.02 ±0.31 mol· mL^-1 to 2.01 ± 0.54 mol · mL^- 1 （ P 〈 0.05 ）. Compared with the model group SOD activity at 24h was significantly raised from 66 ± 1.24 nu/mg pro to 103 ±2.54 nu/mg pro （ P 〈 0.05 ）. Zea Longa law evaluation showed that at 24h degree of neurologic impairment in the acupuncture therapy group was lower than the model group. The score of neurologic impairment had obvious negative correlation with the level of MDA and obvious positive correlation, by contrast, with activity of SOD in brain homogenate. Conclusion In acute stage, acupuncture treatment can protect ischemia brain tissue and lessen the degree of neurologic impairment.
