摘 要:为了有效地进行水浮莲(Pistia stratiotes)的基因工程改造,达到利用水浮莲进行污染水体的植物修复,本研究建立了水浮莲无菌苗培养体系。采用二次消毒法获得水浮莲的无菌苗,并研究了萘乙酸(NAA)、6-苄氨基嘌呤(6-BA)、蔗糖、pH值对其生长的影响,确立了水浮莲的优化培养体系为:1/2 MS+6-BA 0.25mg/L+NAA 0.3mg/L+蔗糖20g/L,pH6.0。水浮莲在此培养基上可快速扩增和长期继代培养,无菌生长的水浮莲比温室中自然水浮莲的植株形态小,易于研究操作。Plants have many endogenous genetic,biochemical and physiological properties that make them ideal tools for soil and water remediation.The genetic engineering plants open up the new possibilities for the phytoremediation.Pistia stratiotes may be a useful plant in remediation of water pollutants by transgenic engineering.Pistia stratiotes is a widely distributed free-floating perennial hydrophyte.Because a large number of organisms are associated with non-axenic Pistia stratiotes,it is important to obtain the axenic culture materials of Pistia stratiotes.So we have developed axenic Pistia cultural system and studied the effects of phytohormone,sucrose and pH on its growth.The results showed that both phytohormone and sucrose have significant effects on the growth of cultured Pistia stratiotes,while pH has little effect.The optimized culture system was determined as following:1/2MS+6-BA 0.25mg/L+NAA 0.3mg/L+ sugar20g/L and pH6.0.The axenic Pistia stratiotes was comparable to the greenhouse-grown plants except for the smaller size.These results will provide a useful basis for genetic transformation of Pistia stratiotes for the remediation of water pollutants.
