摘 要:目的制备莪术油前体脂质体,以期得到不良反应较小的莪术油新剂型。方法采用非水溶剂(叔丁醇-水共溶剂)冷冻干燥法制备莪术油前体脂质体,对莪术油脂质体的包封率、粒径、形态、Zeta电位、溶血性及体外释放速率进行考察。结果莪术油脂质体包封率为92.2%,平均粒径为457nm,Zeta电位为-23.6mV,48h后体外释放大于94.1%,无溶血性,稳定性好。结论采用叔丁醇-水共溶剂冷冻干燥法获得了高包封率、粒径均一、无溶血性、稳定的莪术油前体脂质体。OBJECTIVE To prepare the zedoary turmeric oil (ZTO) proliposomes and develope a new formulation of ZTO with low toxicity and to investigate its properties. METHODS Proliposomes ZTO were prepared by lyophilization tertiary butyl alcohol-water co-solvent system . The properties of the liposomes, including the surface morphology, the size, the entrapment efficiency and the release in vitro, were determined after the profiposomes were changed into the liposomes. RESULTS The liposomes were smooth and spherical. The entrapment efficiency of liposomes ZTO was over 92.2% with an average particle size of 457 nm, the Zeta potential was lower than - 23.6 mV, and the prepared liposomes were stable and safe. After 48 h, 94. 1% of the total drug was released. CONCLUSION The lyophilization methods,using tertiary butyl alcohol-water co-solvent, were availiable for the preparation of ZTO proliposomes with high entrapment efficiency and homogeneous particle size distribution, the prepared liposomes were safe, stable.
