摘  要:目的:建立毛细管区带电泳-电化学检测(CZE-ED)法同时测定夏枯草中芦丁、伞形花内酯、金丝桃苷、对香豆酸、迷迭香酸和咖啡酸6种活性成分的含量。方法:采用自组装的毛细管区带电泳-电化学检测系统,75 cm 长熔融石英毛细管(内径25 μm,外径360 μm),直径300 μm的碳圆盘工作电极,工作电极电位为0.95 V(vs.SCE),分离电压为16 kV,运行液为60 mmol·L^(-1)的硼酸盐缓冲溶液(pH 9.0),进样时间为8 s。结果:6种组分的浓度和峰电流在2~3个数量级范围内呈良好线性关系,最低检测限范围为1.42×10^(-8)~3.75×10^(-7)g·mL^(-1)。结论:该法成功地将夏枯草中的上述6种活性成分进行分离和检测,得到了理想的结果。Objective:To develop a method of capillary zone electrophoresis (CZE) with electrochemical detection (ED) for the determination of six active constituents in Prunella vulgaris L. , namely rutin, umbelliferone, hyperin, p - coumaric acid, rosmarinic acid and caffeic acid. Method: A laboratory - built CZE - ED system and a 75 cm length of 25 μm i. d and 360 μm o. d fused silica capillary was used, a 300 μm diameter carbon disc electrode was employed as the working electrode and the potential was 0. 95 V(vs. SCE). The analytes could be well separated at the separation voltage of 16 kV in a 60 mmol · L^-1 borate buffer( pH 9.0), the injection time was 8 s. Results: Good linear relationships were established between the concentration and peak current of analytes over 2 - 3 orders of magnitude, the detection limits ranged from 1.42 × 10^-8 g · mL^-1to 3.75 × 10^-7 g · mL^- 1. Conclusion:This method has been successfully applied to the determination of active constituents in Prunella vulgaris L. and the assay results are satisfactory.
