摘  要:目的:观察巴戟天正丁醇提取物对乳鼠心肌细胞缺氧/复氧损伤后炎性细胞因子的影响。方法:采用培养的乳鼠心肌细胞建立缺氧/复氧损伤模型,实验分正常细胞培养组、缺氧/复氧损伤模型组、巴戟天正丁醇提物小、中、大剂量组。平衡法测定TNF-α、IL-1β、IL-6的含量变化。结果:巴戟天正丁醇提物可明显改善缺氧、复氧损伤;降低TNF-α、IL-1β、IL-6的含量。结论:巴戟天正丁醇提取物对缺氧/复氧损伤的心肌细胞有防护作用;其防护作用可能与降低TNF-α、IL-1β、IL-6的含量有关。OBJECTIVE To evaluate the effects of the n-butanol extract of Morinda officinalis (BEM) on inflammatory cytokine TNF-α,IL-1β and IL-6 in cultured neonatal rat cardiomyocytes by hypoxia/reoxygenation injury. METHODS The models of hypoxia/reoxygenation were made with th first generation of cultured cardiomyocytes. Cultured cardiomyocytes of neonatal rats were divided into five groups:control group; hypoxia/reoxygenation (H/R) group; the groups of H/R + low dose of BEM(0. 038 g·L^-1), H/R+ medium dose of BEM(0.11 g·L^-1) and H/R+ high dose of (0. 34 g·L^-1 ). The contents of cardiomyocytes TNF-α,IL-1β and IL-6 were tested by balanced technique. RESULTS Group BEM decreased more significantly the TNF-α,IL-1β and IL-6 levels than group model and group control(P〈0. 05). CONCLUSION BEM has obviously protective effect in cultured neonatal rat cardiomyocytes by hypoxia/reoxygenation injury. The protective effect is , to some degree, related to the inhibition of TNF-α,IL-1β and IL-6.
