摘 要:目的应用体外培养的大鼠脑皮层星形胶质细胞,观察抗氧化剂N-乙酰半胱氨酸对糖氧剥夺导致细胞损伤的作用。方法采用第4代大鼠脑皮层星形胶质细胞,以连二亚硫酸钠和无糖Earle液造成化学性糖氧剥夺(OGD)模型,继而恢复完全培养基模拟体内脑缺血再灌注损伤,分别应用MTT法和乳酸脱氢酶(LDH)漏出法检测细胞存活率和细胞溶解程度;应用分光光度法测定星形胶质细胞内乳酸水平和Na+-K+ATP酶活性间接反映脑组织能量代谢变化。结果OGD损伤1 h再恢复24 h后,大鼠皮层星形胶质细胞存活率下降,细胞溶解增加,细胞能量代谢障碍。预先给予N-乙酰半胱氨酸可呈剂量相关性的逆转或减轻细胞损伤。结论N-乙酰半胱氨酸对糖氧剥夺/再恢复所致体外培养的大鼠脑皮层星形胶质细胞损伤具有保护作用。Objective To investigate whether N-acetylcysteine has protective effects on oxygen-glucose deprivation injured cortical astrocytes in rats. Methods The fourth passage of the cortical astrocytes of primary cultured neonate rats was used. Na2S2O4 and sugar free Earle's solution were used to establish the chemical oxygen glucose deprivation (OGD) model, which followed by changing the culture into complete mediums to mimic ischemia/reperfusion !njmy. The 3-(4,5-dimethylthialzol-2-yl)-2-5 -diphe- nyl-tetrazolium bromide (MTT) method and the lactate dehydrogenase (LDH) leakage method were used for evaluations of cell viability and cell lysis, and the Colorimetric method was used to determine changes of cerebral energy metabolism. Results Compared with the sham-group, the cultured neonate rat cortical astrocytes were subjected to one hour of OGD followed by 24 hours of repeffusion, then the cell viability was decreased and the cell lysis was significantly increased, and the cellular energy metabolism was disordered. Pre-treatment with N- acetylcysteine can dose-dependently reverse these injuries. Conclusion N- acetylcysteine exhibits protective effects on OGD/reoxygenation injuries in rat cortical astrocytes.
