摘  要:目的研究宁心红杞胶囊对老龄雌性大鼠卵巢颗粒细胞线粒体以及滑面内质网的影响。方法以自然衰老雌性大鼠为实验模型,以补佳乐(戊酸雌二醇片)为阳性对照药物,按临床等效剂量胃饲宁心红杞胶囊大、中、小剂量8周后,取卵泡颗粒细胞,电镜下观察线粒体以及滑面内质网并摄片,以病理图像分析软件对线粒体、滑面内质网数密度、周密度、面密度指标进行图像分析处理。结果用药后颗粒细胞线粒体和滑面内质网数量和形态上都有所改善,与模型组相比,线粒体及滑面内质网的数密度、周密度、面密度和平均面积都有显著增加(P<0.05)。结论宁心红杞胶囊能够增加卵泡颗粒细胞中线粒体和滑面内质网数量,并改善其形态。Objective To study the effect of Ningxinhongqi Capsules (NC) on microstructure of aged female rats' ovarian granular cells. Methods Fifty natural aged 14-month-old SD rats were equally randomized as five groups and supplied with phytoestrogen-free diet for 8 weeks: menopausal model group with normal diet, positive control group with ethylene estranol (EE) at 0.18 mg · kg^-1 per day, medicine group with heavy dose intake of NC at 0.648 g·kg^-1 per day, middle dose intake of NC at 0.324 g·kg^-1 per day, and small dose intake of NC at 0. 162 g·kg^-1 per day. Ten natural aged 4-month-old SD rats were used as a healthy control group and supplied with phytoestrogen-free food. Then the ovarian granular cells were taken for further observation under the electron microscope and morphometric analysis by using image analysis software . Results Compared with the model group, the indexes including numerical density, perimeter density, area density and average area of mitochondria and smooth endoplasmic reticulum in the medicine group increased significantly (P 〈 0.05). Conclusion Ningxinhongqi Capsules can improve the situation of mitochondria and smooth endoplasmic reticulum both in number and configuration to delay ovarian aging.
