摘　　要：目的:建立以高效液相色谱法测定人血浆中头孢吡肟浓度的方法。方法:色谱柱为DiamonsilC18,流动相为20mmol.L-1醋酸铵溶液(冰醋酸调节pH值至5.00)-乙腈(90:10),流速为1.0mL.min-1,检测波长为254nm,进样量为20μL,柱温为室温。结果:头孢吡肟血药浓度在0.2～50.0μg.mL-1范围内线性关系良好(r=0.9996),定量下限为0.2μg.mL-1;平均方法回收率为99.16%(RSD=1.29%),平均提取回收率为85.43%(RSD=2.37%),日内、日间RSD均<10%。结论:本方法简便、灵敏、快速、准确,可用于人血浆中头孢吡肟的药动学和生物利用度研究。OBJECTIVE： To establish an HPLC method for the determination of cefepime in human plasma. METHODS： The separation of sample was carried on a Diamonsil C18 column with the mobile phase consisted of 20 mmol·L^ -1 ammonium acetate （which was adjusted to pH = 5.00 by acetic acid） - acetonitrile （90 ： 10） at a flow rate of 1.0 mL · min^- 1. The detection wavelength was set at 254 nm; the sample size was 20μL, and the column temperature was set at room temperature. RESULTS： The linear range of cefepime was 0.2-50.0μg · mL^-1 （r = 0.999 6）, with the lower limit of quantitation at 0.2 μg · mL^ -1. The average methodological recovery was 99.16% （RSD = 1.29% ） and the average extraction recovery was 85.43% （RSD = 2.37%） . The inter-day RSD and intraday RSD were all less than 10%. CONCLUSION： The method was simple, sensitive, rapid and accurate and can be used for the pharmacokinetic and bioavailability study of Cefepime.
