摘  要:利用AFLP技术对我国长江南京潜州江段捕获的野生刀鲚(Coilia nasus)资源的遗传多样性进行了分析.筛选出扩增片段多、条带清晰的3对EcoRI/Mse I引物组合,在34尾个体中共扩增出118条清晰的片段,分子量大小在200~1500bp之间,其中多态位点67个,多态性比例为56.78%.种群Nei基因多样性指数为0.2183,Shannon多样性指数为0.3204;种群内遗传距离在0.1263~O.4012之间,显示目前长江刀鲚野生群体的遗传多样性比较丰富.Coilia nasus is a migrating fish shaped like a short knife. The fish swims back from the sea to the river at the beginning of spring, and is considered a special product of the Yangtze River. In recent years, the stocks are depleting and the productivity of natural Coilia nasus from Yangtze River has been reduced. In this study, amplified fragment length polymorphism (AFLP) was used to detect the DNA polymorphism in 34 individuals of natural population of Coilia nasus from Qianzhou, Nanjing Section of Yangtze River. A total of 118 clear, reproducible bands amplified with three optimal AFLP primer combinations were obtained from 34 fish. The length of those bands varied from 200 to 1 500 bp. The percentage of polymorphic fragments was 56. 78%. The average heterozygosity of the population was 0. 218 3 and the index of Shannon diversity was 0. 3204. The genetic distance between individuals was from 0. 126 3 to 0. 401 2. Those results indicated that the genetic diversity of Coilia nasus in Yangtze River was still in a high level. The information may be beneficial to future brood stock selection and defining conservation management for Coilia nasus.
