摘  要:目的:观察电针对SAMP8小鼠海马神经元线粒体超微结构及相关凋亡蛋白的影响,从保护线粒体、抑制细胞凋亡的角度探讨电针治疗老年性痴呆(AD)的部分作用机制。方法:以SAMP8小鼠为研究对象,依据"补肾通督,醒神益智"的治疗原则,电针"百会"、"涌泉",每日1次,共治疗21天。用Morris水迷宫测试小鼠学习记忆能力的变化,评价电针对AD的治疗效应。用透射电镜观察海马神经元线粒体的超微结构,用免疫组化法检测海马神经元中凋亡蛋白Caspase-3、Caspase-9的表达。结果:与空白组小鼠相比较,模型组小鼠平均逃避潜伏期明显延长、原平台象限停留时间明显缩短,海马神经元线粒体超微结构不完整,海马神经元中Caspase-3、Caspase-9表达增高;与模型组小鼠相比较,模针组小鼠治疗后平均逃避潜伏期缩短、原平台象限停留时间延长,学习记忆能力有所提高,海马神经元线粒体超微结构基本正常,海马神经元中Caspase-3、Caspase-9表达减少。结论:电针对AD治疗效应的发挥可能与减轻线粒体超微结构损伤、降低Caspase-3、Caspase-9表达,抑制细胞凋亡线粒体途径的级联反应,减少细胞凋亡有关。Objective :To observe the influence of electro-acupuncture therapy on the mitochondrial ultrastrueture and apoptosis protein in hippoeampal neuron of SAMP8 mice to partly explore the curative mechanism of electro-aeupuncture therapy on Alzheimer's disease(AD) from protecting mitoehondria and inhibiting apoptosis. Methods: SAMP8 mice were acupunctured on"Baihui (GV20) "and" Yongquan( Kidney1 )" according to the therapeutic principle of"Tonifying kidney and activating Govern vessel to reviving brain and promoting intelligence". The treatment was applied once daily and totally lasted for 21 days. 'The changes of learning and memory abilities were observed by Morris water maze to evaluate the curative effect. The mitoehondrial altrastructttres were observe by transmission electron microscope (TEM), and the expressions of apoptosis protein Caspase -3, Caspase -9 of hippoeampal neuron were detect by immunohistoehemistry method. Results : Comparing to normal group, the learning and memory abilities of model group were decreased, the mitoehondrial ultrastructure of hippoeampal neurons were damaged and the expressions of Caspase -3 and Caspase -9 were increased. While comparing to model group, the learning and memory abilities of electro-acupuncture group were promoted, the damages of mitochondrial ultrastructures were relieved, and the expressions of Caspase -3 and Caspase -9 in hippocampal neuron were significantly decreased. Conclusion :The curative mechanism of electro-acupuncture therapy on AD may be related to inhibiting the apoptosis reaction of mitoehondrial route and reducing the apoptosis of hippocampal neuron by relieving the damage of mitechondrial altrastructure and reducing the expressions of Caspase - 3 and Caspase - 9.
