摘 要:目的研究大鼠脑缺血再灌注后热休克蛋白70(HSP70)mRNA的表达,探讨碱性成纤维细胞生长因子(bFGF)对脑组织缺血再灌注神经元HSP70 mRNA的调节作用及机制。方法应用线栓法制作大鼠局灶性脑缺血再灌注模型,大脑中动脉阻塞1h再灌注损伤24h,采用原位缺口末端标记法(TUNEL)、RT-PCR法检测海马及皮质内神经元凋亡和HSP70 mRNA的表达。结果大鼠缺血再灌注海马及顶叶皮质内神经元凋亡数增加而HSP70mRNA表达较正常组增加,注射bFGF后神经元凋亡数减少,HSP70mRNA表达明显高于缺血再灌注组。结论bFGF抑制缺血神经元凋亡,参与HSP70 mRNA的调节,对缺血神经元有保护作用。Objective To investigate the expression of HSP70 mRNA in hippocampus and cortex after cerebral ischemia and reperfusion in rats, and to explore regulative effect and mechanism of basic fibroblast growth factor (bFGF) on HSP70 mRNA in brain tissue. Methods The model of middle cerebral artery occlusion was performed with intraluminal filament blockade. The expression of HSP70 mRNA and cell apoptosis in hippocampus and cortex was detected with RT - PCR method and TUNEL method. Results The expression of HSP70 mRNA in hippocampus and cortex was increased after cerebral ischemia and reperfusion. The expression of HSP70 mRNA in bFGF group was increased; but cell apoptosis was decreased. Conclusion The results indicate that bFGF depress cell apoptosis and participate in the regulation of expression Of HSP70 mRNA in ischemic neurons.
