糖脉平对2型糖尿病大鼠血管中单核细胞趋化蛋白-1表达的影响

摘　　要：目的探讨糖脉平对2型糖尿病大鼠血管中单核细胞趋化蛋白1(MCP-1)表达的影响。方法Wistar大鼠大剂量链脲佐菌素腹腔注射(STZ60 mg/kg),造模前随机分为正常对照组、模型对照组、糖脉平组、二甲双胍组、玉泉丸组,成模后每2周称体重并剪尾测血糖,第6周处死取主动脉标本采用半定量RT-PCR法检测MCP-1mRNA蛋白表达。结果造模后,大鼠血糖明显升高,MCP-1mRNA的表达明显增高,经治疗后,糖脉平治疗组及二甲双胍组MCP-1mRNA的表达明显降低,与糖尿病模型组比较有显著差异(P<0.01)。结论糖脉平能减少糖尿病大鼠MCP-1mRNA的表达,减轻血管内皮局部炎症反应,阻断和降低动脉硬化的形成。Objective To investigate tractant protein-1（MChe expression of blood vessel endothelium monocyte chemoattP-1） in type 2 diabetic rats following Chinese medicine Tang maipingfang.Methods The models of type 2 diabetic Wistar rats were established by injecting high dosage（60mg/kg） of streptozotocin（STZ）,and the successful fifty rats were randomly divided into nommal control group,model control group,Privet Tang maipingfang group,Metformin group and xiaokewan group.Measure the weight and blood sugar by cutting the rats tails ever two weeks.After the sixth weeks execute the rats and take the arteriae maxima Galeni.The expression of MCP-1 was assessed by semiquantitative reverse transcriptase（PCR）.Results After making modle,the blood sugar was obviously rised and the weight was descend.After post-treatment,Comparing the expression of MCP-1,Privet Tang maipingfang group and Metformin group were obviously descend than modle control group（P〈0.01）.Conclusion Privet Tang maipingfang can decrease the expression of MCP-1mRNA levels,abatement inflammatory reaction of blood vessel endothelium,treatment and decrease arteriosclerotic formation.
