摘　　要：目的借助专有软件设计蚋科COI基因的PCR引物,并且检验所设计引物的PCR扩增效率和特异性。方法运用Primer Premier 5.0软件设计4对蚋科COI基因的PCR扩增引物,通过PCR扩增后进行琼脂糖凝胶电泳检测实验。结果在所设计的4对引物中只有1对PCR扩增特异性好且效率高,包含COI基因全序列。结论设计引物时应遵循一定原则,但有一定弹性,可根据模板特点进行设计方法的改良。Objective With the help of Primer Premier 5.0 to design PCR primer about simulium COI gene and verify the efficiency and speciality of PCR about these primers.Methods Four pairs of these primers that would be amplified in simulium COI gene were designed by Primer Premier 5.0,the results of PCR would be investigated through agarose gel electrophoresis.Results One pair of primers in all designed succeeded in the efficiency and specialty of PCR,including COI complete sequence.Conclusion There is the principle of PCR primer design.However,it is flexibility.Based on characteristic of DNA sequence,the means what primer would be designed could be reformed.
