摘  要:目的:探讨心脑舒通胶囊对大鼠局灶性脑缺血再灌注后神经细胞凋亡,及其对炎性因子白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)表达的影响。方法:采用线栓法阻塞大脑中动脉致大鼠局灶性脑缺血再灌注模型,24h后断头取脑,HE染色和尼氏染色光镜下观察大鼠大脑皮层区细胞损伤程度,TUNEL法检测皮层区神经细胞凋亡,ELISA法和放射免疫法分别检测缺血侧脑组织和血清中IL-1β和TNF-α的含量。结果:缺血再灌注模型组神经细胞皮层区明显水肿,组织结构疏松,尼氏小体减少;凋亡细胞数增多;脑组织和血清中IL-1β和TNF-α显著升高,与假手术组比较均有显著性差异(P<0.01)。心脑舒通胶囊能明显减轻皮层区神经细胞水肿,增加尼氏小体,降低细胞凋亡;降低IL-1β和TNF-α含量,与模型组比较,有统计学意义(P<0.05或0.01)。结论:心脑舒通胶囊能减轻缺血再灌注脑组织损伤程度,降低神经细胞凋亡的作用,其机制可能与降低脑组织和血清内IL-1β和TNF-α含量有关。Objective: To study the anti-inflammatory effect of Xinnaoshutong capsule on IL-1β TNF-α and neurocyte apoptosis in cerebral ischemia and reperfusion rats. Methods: Rat models of focal cerebral ischemia and reperfusion were established by thread ligation in middle cerebral artery occlusions. After 24 hours, the brains were removed to observe the morphological changes of rat cerebral cortical neurons under optical microscope through HE staining and Nissel staining, apoptosis of cortical neurons were detected by TUNEL, the content of Interleukin-1β (IL-1β )and tumor necrosis factor-α (TNF-α)in ischemic cerebral brain and serum were detected by ELISA and radioimmunoassay respectively. Results: Compared to control group, ischemia and reperfusion model group have obviously neuronal edema, structure loosing, nissel's body lessening in cerebral cortex and amount of apoptosis neurons. The content of IL-1β and TNF-α increased significantly(P 〈 0.01). Xinnaoshutong capsule could decrease neuronal edema and neuronal apoptosis of cortex, increase the quantity of nissel's body in cortex. And obviously reduce the content of IL-1β and TNF-α in brain tissue and serum(P 〈 0.05 or P〈0.01). Conclusion: Xinnaoshutong capsule has the effect of lightening the injury degree of brain tissues and decreasing neuronal apoptosis after cerebral ischemia and reperfusion. The effect may be related to the decrease of IL- 1β and TNF-α content in brain tissue and serum.
